a drop will connect to the bottom of your frame and go forward so your co2/hpa doesnt stick out so far.

A drop foreward is a fitting you put on a paintball gun so that your air tank is dropped down and forward allowing for a better balance of wieght. Some dont like these because they think it gives them a bigger profile to get hit with.
In this picture, the drop foreward is located on the bottom of the gripframe:

All an expansion chamber will do for you with HPA is act as a gas through grip.

For the reg question, it depents on the gun you use, if you have a stack tube blow back, (spyder, fishy, so on) then you dont need an reg, but if you have a higher end gun (Impy, Cocker, Timmy, so on) then you will need a regulator, which they should have come with.

Not sure what you mean by "gas through grip", but my gun will be a Dragun TES.

Gas through grip is just like a handle, or forgrip, that the gas is routed through from a hose from your tank, into your gun.

As for if you need a reg/expanstion chamber, the answers no, you will be fine without one.
